Anne Stanhope was born circa February 1576.¹ She was the daughter of Sir Thomas Stanhope and Margaret Port.¹ She married Sir John Holles, 1st Earl of Clare , son of Denzill Holles and Eleanor Sheffield , on 23 May 1591 at Shelford, Nottinghamshire, England .¹ She died on 18 November 1651 at Piazza, Covent Garden, London, England .¹ She was buried on 22 December 1651 at St. Mary's Church, Nottingham, Nottinghamshire, England .² Her will (dated 5 November 1651) was probated on 26 November 1651.²
From 23 May 1591, her married name became Holles.¹ As a result of her marriage, Anne Stanhope was styled as Baroness Houghton of Houghton on 9 July 1616. As a result of her marriage, Anne Stanhope was styled as Countess of Clare on 2 November 1624.
